Informatics Inside : Connect(IT) : Informatik-Konferenz an der Hochschule Reutlingen, 9. Mai 2018
(2018)
In the last 20 years there have been major advances in autonomous robotics. In IoT (Industry 4.0), mobile robots require more intuitive interaction possibilities with humans in order to expand its field of applications. This paper describes a user-friendly setup, which enables a person to lead the robot in an unknown environment. The environment has to be perceived by means of sensory input. For realizing a cost and resource efficient Follow Me application we use a single monocular camera as low-cost sensor. For efficient scaling of our Simultaneous Localization and Mapping (SLAM) algorithm, we integrate an inertial measurement unit (IMU) sensor. With the camera input we detect and track a person. We propose combining state of the art deep learning with Convolutional Neural Network (CNN) and SLAM algorithms functionality on the same input camera image. Based on the output robot navigation is possible. This work presents the specification, workflow for an efficient development of the Follow Me application. Our application’s delivered point clouds are also used for surface construction. For demonstration, we use our platform SCITOS G5 equipped with the afore mentioned sensors. Preliminary tests show the system works robustly in the wild.
SLAM systems are mainly applied for robot navigation while research on feasibility for motion planning with SLAM for tasks like bin-picking, is scarce. Accurate 3D reconstruction of objects and environments is important for planning motion and computing optimal gripper pose to grasp objects. In this work, we propose the methods to analyze the accuracy of a 3D environment reconstructed using a LSD-SLAM system with a monocular camera mounted onto the gripper of a collaborative robot. We discuss and propose a solution to the pose space conversion problem. Finally, we present several criteria to analyze the 3D reconstruction accuracy. These could be used as guidelines to improve the accuracy of 3D reconstructions with monocular LSD-SLAM and other SLAM based solutions.
We present a new method for detecting gait disorders according to their stadium using cluster methods for sensor data. 21 healthy and 18 Parkinson subjects performed the time up and go test. The time series were segmented into separate steps. For the analysis the horizontal acceleration measured by a mobile sensor system was considered. We used dynamic time warping and hierarchical custering to distinguish the stadiums. A specificity of 92% was achieved.
Research and Development (R&D) is crucial for the growth and future success of research-based pharma companies. To maintain their R&D organisations efficient, pharmaceutical companies started to hedge the potential of open innovation to cut R&D costs and to access external knowledge. These new strategies could be divided into several categories: open source, innovation centres, crowd sourcing and virtual R&D.
Fitting 3D Morphable Face Models (3DMM) to a 2D face image allows the separation of face shape from skin texture, as well as correction for face expression. However, the recovered 3D face representation is not readily amenable to processing by convolutional neural networks (CNN). We propose a conformal mapping from a 3D mesh to a 2D image, which makes these machine learning tools accessible by 3D face data. Experiments with a CNN based face recognition system designed using the proposed representation have been carried out to validate the advocated approach. The results obtained on standard benchmarking data sets show its promise.
In this exploratory research eight suppliers in the automotive industry are interviewed to measure the application of supply chain finance instruments in their supply chain in the Netherlands and the region of South-West Germany. Current adoption levels and reasons for non adoption are discussed. Based on these indicative results, a set of hypotheses is suggested for further research. The theoretical base of this study is a conceptual model of Supply Chain Finance based on literature research and empirical research in the Netherlands.
Early reduction of risks in a startup or an innovation project is highly important. Appropriate means for risk reduction, such as testing business models with different kinds of experiments exist. However, deciding what to test and how to select the right test, is challenging for many startups and innovation projects. This article presents the so-called Business Experiments Navigator (BEN), a toolkit to assist startup and innovation processes. It compliments other tools such as the Business Model Canvas or the Lean Startup process. The main contribution of BEN is to bridge the gap between the riskiest assumptions of a business model and the multitude of available testing techniques by providing assumption templates. The Business Experiments Navigator has been validated in several workshops. Results show that it creates awareness among the workshop participants that a business model is based on assumptions which impose risks and need to be validated. Further, users of BEN were able to identify relevant assumptions and map different kinds of assumptions to appropriate testing techniques. The process applied in the workshops, as well as the assumption templates, helped the participants understand the main concepts and transfer their learnings, to their own business ideas.
The coculture of osteogenic and angiogenic cells and the resulting paracrine signaling via soluble factors are supposed to be crucial for successfully engineering vascularized bone tissue equivalents. In this study, a coculture system combining primary human adiposederived stem cells (hASCs) and primary human dermal microvascular endothelial cells (HDMECs) within two types of hydrogels based on methacryloyl‐modified gelatin (GM) as three‐dimensional scaffolds was examined for its support of tissue specific cell functions. HDMECs, together with hASCs as supporting cells, were encapsulated in soft GM gels and were indirectly cocultured with hASCs encapsulated in stiffer GM hydrogels additionally containing methacrylate‐modified hyaluronic acid and hydroxyapatite particles. After 14 days, the hASC in the stiffer gels (constituting the “bone gels”) expressed matrix proteins like collagen type I and fibronectin, as well as bone‐specific proteins osteopontin and alkaline phosphatase. After 14 days of coculture with HDMEC‐laden hydrogels, the viscoelastic properties of the bone gels were significantly higher compared with the gels in monoculture. Within the soft vascularization gels, the formed capillary‐like networks were significantly longer after 14 days of coculture than the structures in the control gels. In addition, the stability as well as the complexity of the vascular networks was significantly increased by coculture. We discussed and concluded that osteogenic and angiogenic signals from the culture media as well as from cocultured cell types, and tissue‐specific hydrogel composition all contribute to stimulate the interplay between osteogenesis and angiogenesis in vitro and are a basis for engineering vascularized bone.
This paper investigates the evaluation of dense 3D face reconstruction from a single 2D image in the wild. To this end, we organise a competition that provides a new benchmark dataset that contains 2000 2D facial images of 135 subjects as well as their 3D ground truth face scans. In contrast to previous competitions or challenges, the aim of this new benchmark dataset is to evaluate the accuracy of a 3D dense face reconstruction algorithm using real, accurate and high-resolution 3D ground truth face scans. In addition to the dataset, we provide a standard protocol as well as a Python script for the evaluation. Last, we report the results obtained by three state-of-the-art 3D face reconstruction systems on the new benchmark dataset. The competition is organised along with the 2018 13th IEEE Conference on Automatic Face & Gesture Recognition.
An interactive clothing design and a personalized virtual display with user’s own face are presented in this paper to meet the requirement of personalized clothing customization. A customer interactive clothing design approach based on genetic engineering ideas is analyzed by taking suit as an example. Thus, customers could rearrange the clothing style elements, chose available color, fabric and come up with their own personalized suit style. A web 3D customization prototype system of personalized clothing is developed based on the Unity3D and VR technology. The layout of the structure and functions combined with the flow of the system are given. Practical issues such as 3D face scanning, suit style design, fabric selection, and accessory choices are addressed also. Tests to the prototype system indicate that it could show realistic clothing and fabric effect and offer effective visual and customization experience to users.
Completely defined co-culture of adipogenic differentiated ASCs and microvascular endothelial cells
(2018)
Vascularized adipose tissue models are in high demand as alternatives to animal models to elucidate the mechanisms of widespread diseases, screen for new drugs or assess drug safety levels. Animal-derived sera such as fetal bovine serum (FBS), which are commonly used in these models, are associated with ethical concerns, risk of contaminations and inconsistencies of their composition and impact on cells. In this study, we developed a serum-free, defined co culture medium and implemented it in an adipocyte/endothelial cell (EC) co culture model.
Human adipose-derived stem cells were differentiated under defined conditions (diffASCs) and, like human microvascular ECs (mvECs), cultured in a defined co culture medium in mono-, indirect or direct co-culture for 14 days. The defined co-culture medium was superior when compared to mono-culture media and facilitated the functional maintenance and maturation of diffASCs including perilipin A expression, lipid accumulation, and also glycerol and leptin release. The medium also allowed mvEC maintenance, confirmed by the expression of CD31 and von Willebrand factor (vWF), and by acetylated low density lipoprotein (acLDL) uptake. Thereby, mvECs showed strong dependence on EC-specific factors. Additionally, mvECs formed vascular structures in direct co-culture with diffASCs.
The completely defined co-culture system allows for the serum-free culture of adipocyte/EC co-cultures and thereby represents a valuable and ethically acceptable tool for the culture and study of vascularized adipose tissue models.
To date, special interest has been paid to composite scaffolds based on polymers enriched with hydroxyapatite (HA). However, the role of HA containing different trace elements such as silicate in the structure of a polymer scaffold has not yet been fully explored. Here, we report the potential use of silicate-containing hydroxyapatite (SiHA) microparticles and microparticle aggregates in the predominant range from 2.23 to 12.40 μm in combination with polycaprolactone (PCL) as a hybrid scaffold with randomly oriented and well-aligned microfibers for regeneration of bone tissue. Chemical and mechanical properties of the developed 3D scaffolds were investigated with XRD, FTIR, EDX and tensile testing. Furthermore, the internal structure and surface morphology of the scaffolds were analyzed using synchrotron X-ray μCT and SEM. Upon culturing human mesenchymal stem cells (hMSC) on PCL-SiHA scaffolds, we found that both SiHA inclusion and microfiber orientation affected cell adhesion. The best hMSCs viability was revealed at 10 day for the PCL-SiHA scaffolds with well-aligned structure (~82%). It is expected that novel hybrid scaffolds of PCL will improve tissue ingrowth in vivo due to hydrophilic SiHA microparticles in combination with randomly oriented and well-aligned PCL microfibers, which mimic the structure of extracellular matrix of bone tissue.
Bone remodeling can be mimicked in vitro by co-culture models. Based on bone cells, such co-cultures help to study synergistic morphological changes and the impact of materials and applied substances. Hence, we examined the formation of osteoclasts on bovine bone materials to prove the bone resorption functionality of the osteoclasts in three different co-culture set-ups using human monocytes (hMCs) and (I) human mesenchymal stem cells (hMSCs), (II) osteogenic differentiated hMSCs (hOBs), and (III) hOBs in addition of soluble monocyte-colony stimulating factor (M CSF) and cytokine receptor activator of NFkB ligand (RANKL).We detected osteoclast-specific actin morphology, as well as the expression of cathepsin K and CD51/61 in single cells in set-up II and in numerous cells in set-up III. Resorption pits on bone material as characteristic proof of functional osteoclasts were not found in set-up I and II, but we detected such resorption pits in set–up III. We conclude in co culture models without M-CSF and RANKL that monocytes can differentiate into osteoclasts that show the characteristic actin structures and protein expression. However, to receive functional bone resorbing osteoclasts in vitro, the addition of M-CSF and RANKL is needed. Moreover, we suggest the use of bone or bone-like materials for future studies evaluating osteoclastogenesis.
Nowadays robust, energy-efficient multisensor microsystems often come with heavily restricted power budgets and the characteristic of remaining in certain states for a longer period of time. During this time frame there is no continuous clock signal required which gives the opportunity to suspend the clock until a new transition is requested. In this paper, we present a new topology for on-demand locally clocked finite state machines. The architecture combines a local adaptive clocking approach with synchronous and asynchronous components forming a quasi synchronous system. Using adaptive and local clocking comes with the advantages of reducing the power consumption while saving design effort when no global clock tree is needed. Combining synchronous and asynchronous components is beneficial compared to previous fully asynchronous approaches concerning the design restrictions. The developed topology is verified by the implementation and simulation of a temperature-ADC sensor system realized in a 180 nm process.

A transaction is a demarcated sequence of application operations, for which the following properties are guaranteed by the underlying transaction processing system (TPS): atomicity, consistency, isolation, and durability (ACID). Transactions are therefore a general abstraction, provided by TPS that simplifies application development by relieving transactional applications from the burden of concurrency and failure handling. Apart from the ACID properties, a TPS must guarantee high and robust performance (high transactional throughput and low response times), high reliability (no data loss, ability to recover last consistent state, fault tolerance), and high availability (infrequent outages, short recovery times).
The architectures and workhorse algorithms of a high-performance TPS are built around the properties of the underlying hardware. The introduction of nonvolatile memories (NVM) as novel storage technology opens an entire new problem space, with the need to revise aspects such as the virtual memory hierarchy, storage management and data placement, access paths, and indexing. NVM are also referred to as storage-class memory (SCM).
Active storage
(2018)
In brief, Active Storage refers to an architectural hardware and software paradigm, based on collocation storage and compute units. Ideally, it will allow to execute application-defined data ... within the physical data storage. Thus Active Storage seeks to minimize expensive data movement, improving performance, scalability, and resource efficiency. The effective use of Active Storage mandates new architectures, algorithms, interfaces, and development toolchains.
Blockchains yield to new workloads in database management systems and K/V-stores. Distributed Ledger Technology (DLT) is a technique for managing transactions in ’trustless’ distributed systems. Yet, clients of nodes in blockchain networks are backed by ’trustworthy’ K/V-Stores, like LevelDB or RocksDB in Ethereum, which are based on Log-Structured Merge Trees (LSM Trees). However, LSM-Trees do not fully match the properties of blockchains and enterprise workloads.
In this paper, we claim that Partitioned B-Trees (PBT) fit the properties of this DLT: uniformly distributed hash keys, immutability, consensus, invalid blocks, unspent and off-chain transactions, reorganization and data state / version ordering in a distributed log-structure. PBT can locate records of newly inserted key-value pairs, as well as data of unspent transactions, in separate partitions in main memory. Once several blocks acquire consensus, PBTs evict a whole partition, which becomes immutable, to secondary storage. This behavior minimizes write amplification and enables a beneficial sequential write pattern on modern hardware. Furthermore, DLT implicate some type of log-based versioning. PBTs can serve as MV-store for data storage of logical blocks and indexing in multi-version concurrency control (MVCC) transaction processing.
Modern persistent Key/Value stores are designed to meet the demand for high transactional throughput and high data ingestion rates. Still, they rely on backwards-compatible storage stack and abstractions to ease space management, foster seamless proliferation and system integration. Their dependence on the traditional I/O stack has negative impact on performance, causes unacceptably high write-amplification, and limits the storage longevity.
In the present paper we present NoFTL KV, an approach that results in a lean I/O stack, integrating physical storage management natively in the Key/Value store. NoFTL-KV eliminates backwards compatibility, allowing the Key/Value store to directly consume the characteristics of modern storage technologies. NoFTLKV is implemented under RocksDB. The performance evaluation under LinkBench shows that NoFTL-KV improves transactional throughput by 33%, while response times improve up to 2.3x. Furthermore, NoFTL KV reduces write-amplification 19x and improves storage longevity by imately the same factor.
Background: Internationally, teledermatology has proven to be a viable alternative to conventional physical referrals. Travel cost and referral times are reduced while patient safety is preserved. Especially patients from rural areas benefit from this healthcare innovation. Despite these established facts and positive experiences from EU neighboring countries like the Netherlands or the United Kingdom, Germany has not yet implemented store-and-forward teledermatology in routine care.
Methods: The TeleDerm study will implement and evaluate store-and-forward teledermatology in 50 general practitioner (GP) practices as an alternative to conventional referrals. TeleDerm aims to confirm that the possibility of store-and-forward teledermatology in GP practices is going to lead to a 15% (n = 260) reduction in referrals in the intervention arm. The study uses a cluster-randomized controlled trial design. Randomization is planned for the cluster “county”. The main observational unit is the GP practice. Poisson distribution of referrals is assumed. The evaluation of secondary outcomes like acceptance, enablers and barriers uses a mixed methods design with questionnaires and interviews.
Discussion: Due to the heterogeneity of GP practice organization, patient management software, information technology service providers, GP personal technical affinity and training, we expect several challenges in implementing teledermatology in German GP routine care. Therefore, we plan to recruit 30% more GPs than required by the power calculation. The implementation design and accompanying evaluation is expected to deliver vital insights into the specifics of implementing telemedicine in German routine care.
We present an approach for segmenting individual cells and lamellipodia in epithelial cell clusters using fully convolutional neural networks. The method will set the basis for measuring cell cluster dynamics and expansion to improve the investigation of collective cell migration phenomena. The fully learning-based front-end avoids classical feature engineering, yet the network architecture needs to be designed carefully. Our network predicts how likely each pixel belongs to one of the classes and, thus, is able to segment the image. Besides characterizing segmentation performance, we discuss how the network will be further employed.
This work is a report on practical experiences with the issue of interoperability in German practice management systems (PMSs) from an ongoing clinical trial on teledermatology, the TeleDerm project. A proprietary and established web-platform for store-and-forward telemedicine is integrated with the IT in the GPs’ offices for automatic exchange of basic patient data. Most of the 19 different PMSs included in the study sample lack support of modern health data exchange standards, therefore the relatively old but widely available German health data exchange interface “Gerätedatentransfer” (GDT) is used. Due to the lack of enforcement and regulation of the GDT standard, several obstacles to interoperability are encountered. As a partial, but reusable working solution to cope with these issues, we present a custom middleware which is used in conjunction with GDT. We describe the design, technical implementation and observed hindrances with the existing infrastructure. A discussion on health care interfacing standards and the current state of interoperability in German PMS software is given.
Properties data of phenolic resins synthetized for the impregnation of saturating Kraft paper
(2018)
The quality of decorative laminates boards depends on the impregnation process of Kraft papers with a phenolic resin, which constitute the raw materials for the manufacture of the cores of such boards. In the laminates industries, the properties of resins are adapted via their syntheses, usually by mixing phenol and formaldehyde in a batch, where additives, temperature and stirring parameters can be controlled. Therefore, many possibilities of preparation and phenolic resins exist, that leads to different combinations of physico chemical properties. In this article, the properties data of eight phenolic resins synthetized with different parameters of pH and reaction times at 60 °C and 90 °C are presented: the losses of pH after synthesis and the dynamic viscosities measured after synthesis and one the solid content is adjusted to 45%w/w in methanol. Data aquired by Differential Scanning Calorimetry (DSC) of the resins and Inverse Gas Chromatography (IGC) of cured solids are given as well.
The current paper studies the influence of geometrical parameters of the fused deposition modeling (FDM) - fused filament fabrication (FFF) 3D printing process on printed part strength for open source desktop 3D printers and the most popular material used for that purpose - i.e., polylactic acid (PLA). The study was conducted using a set of different nozzles (0.4, 0.6, and 0.8 mm) and a range of layer heights from the minimum to maximum physical limits of the machine. To assess print strength, a novel assessment method is proposed. A tubular sample is loaded in the weakest direction (across layers) in a three point bending fixture. Mesostructure evaluation through scanning electronic microscopy (SEM) scans of the samples was used to explain the obtained results. We detected a significant influence of geometric process parameters on sample mesostructure, and consequently, on sample strength.
Lots of movies are produced every year, too many to watch all of them and in particular, to get an overview about the evolution of typical movie genres and actors playing in them. Moreover, it is a challenging problem to detect correlations among the movies and the actors in those movies, in particular, if we are interested in time-varying data patterns like trends, countertrends, or anomalies and outliers. Those correlations are specifically interesting if they can be inspected on different levels of granularity, e.g., temporal, but also hierarchical in form of country- or continent-based correlations. In this paper we describe the IMDb Explorer, a webbased visualization tool that consists of two major views denoted by the movie cosmos and the career lines. Both views are linked and interactively manipulable while a list of user-defined metrics are explorable. We illustrate the usefulness of the visualization tool by applying it to the entire movie database provided by IMDb.

The topics of the tutorials were The human factor in agile transitions – using the personas concept in agile oaching, Process Management 4.0 – Best Practices, and Domain-specific languages for specification, development, and testing of autonomous systems.
Being able to monitor the heart activity of patients during their daily life in a reliable, comfortable and affordable way is one main goal of the personalized medicine. Current wearable solutions lack either on the wearing comfort, the quality and type of the data provided or the price of the device. This paper shows the development of a Textile Sensor Platform (TSP) in the form of an electrocardiogram (ECG)-measuring T-shirt that is able to transmit the ECG signal to a smartphone. The development process includes the selection of the materials, the design of the textile electrodes taking into consideration their electrical characteristics and ergonomy, the integration of the electrodes on the garment and their connection with the embedded electronic part. The TSP is able to transmit a real-time streaming of the ECG-signal to an Android smartphone through Bluetooth Low Energy (BLE). Initial results show a good electrical quality in the textile electrodes and promising results in the capture and transmission of the ECG signal. This is still a working- progress and it is the result of an interdisciplinary master project between the School of Informatics and the School of Textiles & Design of the Reutlingen University.
A series of novel biomedical TPCUs with different percentages of hard segment and a silicone component in the soft segment were synthesized in a multi stage one-pot method. The kinetic profiles of the urethane formation in TPCU-based copolymer systems were monitored by rheological, in line FTIR spectroscopic (React IR) and real-time calorimetric (RC1) methods. This process-analytically monitored multi step synthesis was successfully used to optimize the production of medical-grade TPCU elastomers on preparative scale (in lots of several kg) with controlled molecular structure and mechanical properties. Various surface and bulk analytical methods as well as systematic studies of the mechanic response of the elastomer end-products towards compression and tensile loading were used to estimate the bio-stability of the prepared TPCUs in vitro after 3 months. The tests suggested that high bio-stability of all polyurethane formulations using accelerating in vitro test can be attributed to the synthetic design as well as to the specific techniques used for specimen preparation, namely: (1) the annealing for reducing residual polymer surface stress and preventing IES, (2) stabilization of the morphology by long time storage of the specimens after processing before being immersed in the test liquids, (3) purification by extraction to remove the shot chain oligomers which are the most susceptible to degradation. All mechanical tests were performed on cylindrical and circular disc specimens for modelling the thickness of the meniscus implants under application-relevant stress conditions.
So far, only few authors addressed the serum-free, defined differentiation of adipocytes. And there are hardly any trials available on the defined maintenance of adipocytes. In this study, the development of a defined culture medium for the adipogenic differentiation of primary human adipose-derived stem cells (ASCs) was aimed. Based on the addition of specific factors for the replacement of serum, ASCs were differentiated to viable and characteristic adipocytes for 14 days, which was proven through the accumulation of lipids, the expression of perilipin A and by the release of leptin and glycerol. Furthermore, a defined maintenance medium was developed, which supported the maturation and stability of cells for a long-term period of additional 42 days until day 56.
This paper addresses what we call the investment question: under what plausible circumstances, if any, can variable renewable energy (VRE, and solar photovoltaic (PV) in particular) be a good investment? Although VRE has been growing rapidly world-wide, it is generally subsidized. Under what cost and market conditions can solar PV flourish without subsidy? We employ solar insolation and market price data from the U.S. and from Germany to gain insight into the investment question. We find that unsubsidized solar PV is or may soon be a justifiable investment, but that market arrangements may play a crucial role in determining success. We end by sketching a proposal that amounts to a reformed capacity market that would afford participation of solar PV.
The focus of the developed maturity model was set on processes. The concept of the widespread CMM and its practices has been transferred to the perioperative domain and the concept of the new maturity model. Additional optimization goals and technological as well as networking-specific aspects enable a process- and object-focused view of the maturity model in order to ensure broad coverage of different subareas. The evaluation showed that the model is applicable to the perioperative field. Adjustments and extensions of the maturity model are future steps to improve the rating and classification of the new maturity model.
A shift in attitudes to purchasing departments can be perceived. No longer is the chief goal solely to reduce costs; the procurement function is assuming strategic relevance in the business model, leveraging the supplier as a foundation for innovation. The knowledge accumulated by suppliers is accessed over the journey of long-term partnerships to streamline business practice. Businesses are finding themselves in increasingly competitive environments, and thus need to address inefficiencies in supplier management. “Procurement 4.0” is a concept used in discussing digitalisation in business processes, referring to the process of supplier relationship management and optimisation. This model and its application to supplier relationship management will be the focus of this article. Realising the efficiencies to be obtained in buyer-supplier relationships through “Procurement 4.0” will be explored, primarily through an emphasis on digitalisation of the relationship between the procurement department and supplier.
Surface topographies are often discussed as an important parameter influencing basic cell behavior. Whereas most in vitro studies deal with microstructures with sharp edges, smooth, curved microscale topographies might be more relevant concerning in-vivo situations. Addressing the lack of highly defined surfaces with varying curvature, we present a topography chip system with 3D curved features of varying spacing, curvature radii as well as varying overall dimensions of curved surfaces. The CurvChip is produced by low-cost photolithography with thermal reflow, subsequent (repetitive) PDMS molding and hot embossing. The platform facilitates the systematic in-vitro investigation of the impact of substrate curvature on cell types like epithelial, endothelial, smooth muscle cells, or stem cells. Such investigations will not only help to further understand the mechanism of curvature sensation but may also contribute to optimize cell-material interactions in the field of regenerative medicine.
In Neurofibromatosis 1 (NF1) germ line loss of function mutations result in reduction of cellular neurofibromin content (NF1+/−, NF1 haploinsufficiency). The Ras-GAP neurofibromin is a very large cytoplasmic protein (2818 AA, 319 kDa) involved in the RAS-MAPK pathway. Aside from regulation of proliferation, it is involved in mechanosensoric of cells. We investigated neurofibromin replacement in cultured human fibroblasts showing reduced amount of neurofibromin. Full length neurofibromin was produced recombinantly in insect cells and purified. Protein transduction into cultured fibroblasts was performed employing cell penetrating peptides along with photochemical internalization. This combination of transduction strategies ensures the intracellular uptake and the translocation to the cytoplasm of neurofibromin. The transduced neurofibromin is functional, indicated by functional rescue of reduced mechanosensoric blindness and reduced RasGAP activity in cultured fibroblasts of NF1 patients or normal fibroblasts treated by NF1 siRNA. Our study shows that recombinant neurofibromin is able to revert cellular effects of NF1 haploinsuffiency in vitro, indicating a use of protein transduction into cells as a potential treatment strategy for the monogenic disease NF1.
